Despite the large number of GPCRs and their ligands, intracellular signal transduction after GPCRs binding to ligands is mainly achieved by four G-protein families (Gαs, Gαi/Gα0, Gαq/Gα11, and Gα12/Gα13). G proteins are heterotrimers, consisting of three distinct subunits: an α subunit, a β subunit, and a γ subunit. GPCRs signal transduction mechanisms are mainly divided into three categories: Gq(phospholipase C, IP3/IP1/Ca2+), Gi and Gs(inhibiting and stimulating adenylate cyclase, respectively, and regulating cAMP production). Quantitative determination of intracellular cAMP, IP3/IP1/Ca2+ can be used to evaluate the titer of the drug on the activation or inhibition of GPCR.
