GPCRs are transmembrane proteins constitute a superfamily of genes that involved in signal transduction. Research has shown that GPCRs play an important role in the treatment of high blood pressure, neurological diseases, pain, and other conditions. Despite the large number of GPCRs and their ligands, intracellular signal transduction after GPCRs binding to ligands is mainly achieved by four G-protein families (Gαs, Gαi/Gα0, Gαq/Gα11, and Gα12/Gα13). G proteins are heterotrimers, consisting of three distinct subunits: an α subunit, a β subunit, and a γ subunit. GPCRs signal transduction mechanisms are mainly divided into three categories: Gq(phospholipase C, IP3/IP1/Ca2+), Gi and Gs(inhibiting and stimulating adenylate cyclase, respectively, and regulating cAMP production). Quantitative determination of intracellular cAMP, IP3/IP1/Ca2+ can be used to evaluate the titer of the drug on the activation or inhibition of GPCR.
